LifeCyte offers comprehensive and quality services in protein production, purification and characterization utilizing both traditional and progressive biochemical technologies.

  • Protein Production
    • LifeCyte provides professional consultation to optimize the expression and production needs of both native and recombinant proteins, either secreted or intracellular from a variety of sources.  We are experienced in vector design optimization and construction (see Molecular Biology) as well as viral transduction and vector delivery into a suitable cell line or host cell (bacterial, mammalian or insect/baculovirus) (see Cell Biology).  The formulation of stable cell lines expressing the protein of interest can also be provided as an additional resource.  Customized tagged proteins (FLAG, GFP, HA, etc.) can be engineered per request for subsequent use in localization studies (see Cell Biology), or pull-down (immuno-precipitation) assays used in immuno-purification reactions (see Immunology).
  • Protein Purification
    • Specific strategies can be discussed with the customer to ascertain the most desirable method to purify proteins to homogeneity.  We can optimize conditions for detergent solubility for extraction from membranes, and develop lysis conditions for protease and phosphatase-free intracellular protein (both whole-cell and nuclear) extraction.  LifeCyte offers an extensive array of chromatographic, electrophoretic and immunological technologies for purification of the protein of interest [Affinity purification, Ion- and Size exclusion chromatography, HPLC, FPLC].  Spectrophotometric enzyme assays can also be employed as a means to monitor the protein purification during the production run.
  • Protein Analysis/Characterization
    • Protein analysis studies are employed to add quality assurance for the detection, identification and characterization of the final high quality protein product.  Resources for protein analysis include protein concentration assays, electrophoresis [SDS-PAGE, 2D gel electrophoresis, nondenaturation gel electrophoresis, isoelectric focusing (IEF), electrophoretic mobility shift assay (EMSA),] coupled with Western blot and thin-layer chromatographic analysis, sedimentation gradient fractionation, immunoprecipitation and mass spectroscopic analysis [LC/MS, LC/MS/MS, ESI, MS, MALDI-TOF]. 
  • Additional core biochemistry methods can also be employed depending upon the need including centrifugation [ultra, gradient, etc.], Kd analysis, equilibrium dialysis, and solubility analysis assays.
